they're trying to cure aging and actually have a serious, comprehensive plan to do it with many preftigious ppl on board

SENS actually got some bitcoin $$$ recently too but basically has an unlimited need for cash since they're doing basic scientific research on so many problems

senescent cell removal by itself extends lifespan in mice by up to 35%. there are several senolytic candidate drugs now that could possibly extend median lifespan by 1-3 decades. drugs like dasatinib are not something the average boomer is going to want to take without human trials, but it seems likely that relatively benign senolytics will be available in less than 10 years.

SENS is likely taking the wrong path. The induced pluripotent stem cell research should be taken as strong evidence that epigenetic changes are the primary cause of aging. the research being done on cyclic use of reprogramming factors to undo epigenetic changes (which isn't part of the SENS research project) is more likely to lead to large increases in maximum lifespan than anything in the SENS program.

Feinerman: There is growing evidence that epigenetic changes are highly organized and may be one of the causes of ageing. What do you think? Maybe should we consider epigenetic changes as another type of damage in SENS model, calling EpiSENS?

de Grey: We need to be much more precise with definitions in order to answer your question. Epigenetic changes can be classified into two main classes: shift and noise. Shift means changes that occur in a coordinated manner among all cells of a given type and tissue, whereas noise means changes that occur in some such cells but not others, increasing the variability of that type of cell. Shifts are caused by some sort of program (genetic changes to the cell's environment), so yes, they can potentially be reversed by restoring the environment and putting the program into reverse. Noise, on the other hand, is not reversible. And we have for several years worked on determining whether it happens enough to matter in a currently normal lifetime. We have not got to a definitive answer, but it's looking though no, epigenetic noise accumulates too slowly to matter, other than maybe for cancer (which, of course, we are addressing in other ways).

Feinerman: Should we use reprogramming factors to reverse the epigenetic program?

de Grey: Probably not. There may be some benefits in doing so, as a way to restore the numbers of certain types of stem cells, but we can always do that by other methods (especially by direct stem cell transplantation), so I don't think we will ever actually need to dedifferentiate cells in vivo.

I agree with him that stochastic epigenetic drift is probably not very important (like stochastic nuclear DNA damage doesn't matter in a normal human lifespan). There are, however, highly regular epigenetic changes that are strongly predictive of mortality. Old cells that are reprogrammed show almost complete reversal of the aged phenotype. Aging is mostly programmed.

His concern with in vivo reprogramming is probably related to cancer (if cells completely lose their identity cancer formation is probable). The study done using temporary application of cellular reprogramming factors suggests this isn't necessarily a major problem.

MOUNTAIN VIEW, Calif., Jan. 16, 2018 (GLOBE NEWSWIRE) -- SENS Research Foundation (SRF), a leading Silicon Valley nonprofit focused on diseases of aging, announced today that it received over $5 Million in donations during its year end fundraising campaign. These donations included $1 Million in Bitcoin from the Pineapple Fund; $1 Million in Bitcoin from an anonymous donor; and $2.4 Million in Ethereum from Vitalik Buterin, the cofounder of Ethereum and Bitcoin Magazine.

“SENS Research Foundation is pleased by the strong support we have received from members of the tech community who are innovative leaders in utilizing cryptocurrency. We appreciate their support and look forward to partnering with them going forward,” said Aubrey de Grey, CSO.

“We are very grateful to all of our donors for their incredible support of our Year End Campaign,” said Jerri Barrett, VP of Outreach. “Our initial campaign goal was $250,000. We were thrilled to receive over 1400 donations totaling over $5 million in just ten weeks.”

“Achieving this level of donation in such a short period of time shows that the momentum SENS Research Foundation has achieved is continuing,” said Michael Kope, CEO. “We are looking forward to engaging even more of the tech community in our work and to continue to accelerate our progress through the expansion of our research programs. Their support makes this growth possible.”

Aubrey has talked about this on various occasions. One issue is the wife often hates it. So like a handful of dumb shrews who'd rather give to causes that get them more cocktail party status points will be responsible for significantly more deaths than Adolf Hitler

i expect this will change once people start seeing robust rejuvenation from senolytics. the idea of curing or substantially reversing aging still sounds fanciful to most.
