"There is an SNP (RS4680) which is thought to be a genetic basis for modafinil non-response. This gene data is included in 23andme's genotyping service for those that have had it done. About 25% of people of caucasion decent are (A,A) at rs4680, which is in the COMT gene whose enzyme degrades dopamine/epinephrine. Modafinil probably works by raising these catecholamines. GG carriers respond quite well to 200mg modafinil, while AA carriers barely respond at all. From anectodal experience AG carriers (myself) respond well to modafinil. I have unknown personal data about GG carriers.
Codes to help you decipher any literature you may come across:
Met/Met=AA; High dopamine in prefrontal cortex. Val/Met=AG; Higher dopamine in prefrontal cortex (not as high as AA). Val/Val= GG; Lower dopamine in prefrontal cortex. It looks like AG is between GG and AA. If this SNP is highly correlatory to modafinil response then this theory should hold true: GG respond very well, AG respond ok, AA dont respond well if at all."
