The FDA approved drug rapamycin increases lifespan in rodents and delays age-related dysfunction in rodents and humans. Nevertheless, important questions remain regarding the optimal dose, duration, and mechanisms of action in the context of healthy aging. Here we show that 3 months of rapamycin treatment is sufficient to increase life expectancy by up to 60% and improve measures of healthspan in middle-aged mice.

Metformin produces inconsistent effects in rodents. NMN or NR are interesting put still fairly unproven. If i had to guess, both probably work to some extent in humans, although the effects are unlikely to be large

I think there is a high probability rapamycin works in humans. It works in human cell cultures and it consistently works in a wide range of species. it seems to be synergistic with metformin
